MOTS-C (Mitochondrial Open Reading Frame of the 12S rRNA-c) is a 16-amino acid mitochondrial-derived peptide encoded within mitochondrial DNA. Studied as a mitochondrial signalling molecule, MOTS-C is one of the most closely watched peptides in metabolic health and longevity research. How It WorksMOTS-C acts as an intracellular signalling molecule, activating the AMPK pathway — a key regulator of cellular energy balance. Research indicates it supports glucose regulation, mitochondrial function, and the body's adaptive response to metabolic stress and ageing. What is MOTS-C Pen?
MOTS-c is a short peptide encoded in mitochondrial DNA, within the gene for 12S ribosomal RNA. It was first described in 2015 by researchers at the University of Southern California. It belongs to a group known as mitochondrial-derived peptides.
MOTS-c is thought to act partly through the AMPK pathway, which helps cells sense and manage energy. Research suggests it can move into the cell nucleus during metabolic stress and influence gene activity. Levels in the body appear to change with age and exercise.

What the research covers
MOTS-c has been studied mainly in cells and animals. In the original 2015 study, mice given MOTS-c showed resistance to diet-induced weight gain and improved insulin sensitivity. Later animal studies have looked at exercise capacity, ageing and muscle metabolism. Human research so far is largely observational, measuring natural MOTS-c levels in relation to insulin sensitivity and other markers. Controlled human trials of MOTS-c as a treatment are very limited. MOTS-c is not approved as a medicine by the US FDA.

Frequently asked questions
What makes MOTS-c different from other peptides?
Most peptides are encoded by DNA in the cell nucleus. MOTS-c is unusual because it is encoded by DNA inside mitochondria, the energy-producing parts of cells.
What does the research show?
Animal studies have looked at insulin sensitivity, weight gain and exercise capacity. Human studies are mostly observational, and controlled treatment trials are limited.
What formats are available and how are they stored?
It is available as a pen, cartridge or vial. Keep pens and cartridges in the fridge, and refrigerate vials after reconstitution, away from light. Follow the storage instructions on your pharmacy label.
Are there side effects?
Side effects can occur. Your GP will review your health, medication and, where relevant, blood results before prescribing, and you should report anything unexpected.
How much should I take?
Your dose is set by our licensed GP following a consultation, and printed on your pharmacy label.
