GLP-3

Retatrutide is a novel triple agonist peptide studied for human-relevant research on metabolic regulation, appetite control, and metabolic regulation. It combines GLP-1, GIP, and glucagon receptor activity to provide a comprehensive approach for investigating glycemic control, energy balance, and cardiometabolic health. Key Features cGMP manufactured in an FDA-registered facility ≥98% purity confirmed by HPLC analysis Batch-specific Certificates of Analysis (COAs) included Human-Relevant Research Applications Glycemic control and insulin sensitivityImproves glycemic markers (HbA1c, fasting glucose) in type 2 diabetes and metabolic syndrome contexts.https://pubmed.ncbi.nlm.nih.gov/40291085/ Metabolic regulation and appetite regulationLeads to significant metabolic regulation and appetite signalling modulation.https://pubmed.ncbi.nlm.nih.gov/40916752/ Cardiometabolic researchAssociated with better metabolic outcomes including weight, glucose, BMI, and blood pressure in clinical trials.https://pubmed.ncbi.nlm.nih.gov/39318607/ Mechanism Summary Dual and triple receptor activity (GLP-1, GIP, glucagon) enhances glucose regulation and metabolic outcomes. Modulates satiety and energy homeostasis, supporting human-relevant research in obesity and diabetes. Enables studies on cardiometabolic health, metabolic regulation, and insulin sensitivity. Frequently Research Questions How does Retatrutide differ from GLP-1 analogues?Retatrutide's triple receptor activity offers broader metabolic and metabolic regulation effects than GLP-1 alone. Is it suitable for long-term human-relevant studies?Yes. It allows chronic investigation of insulin sensitivity, weight control, and cardiometabolic outcomes. Does it affect cardiovascular markers?Studies show beneficial effects on lipid profiles, blood pressure, and other cardiometabolic indicators. Scientific References Frias et al. What is GLP-3?

Retatrutide is an investigational synthetic peptide developed by Eli Lilly under the code LY3437943. It is sometimes called a 'GLP-3' in consumer settings, but this is not a scientific name. It is a single molecule designed to activate three hormone receptors: GIP, GLP-1 and glucagon.

The peptide carries a fatty acid side chain that helps it bind to albumin in the blood, which extends how long it stays in the body. GIP and GLP-1 activity is thought to influence appetite and insulin release. Glucagon receptor activity is thought to increase energy expenditure and influence liver fat.

Also known as: Retatrutide, LY3437943, GLP-3 RX, Triple agonist, GGG tri-agonist

What are peptides?

What the research covers

Retatrutide has been studied in randomised, placebo-controlled human trials. A phase 2 trial in adults with obesity, published in the New England Journal of Medicine in 2023, reported dose-dependent reductions in body weight over 48 weeks. Phase 2 work has also looked at liver fat and type 2 diabetes. Larger phase 3 trials are under way, and gastrointestinal effects such as nausea were the most commonly reported side effects in early studies. Long-term safety data are still being collected. Retatrutide is an investigational medicine and is not approved by the US FDA or other major regulators.

Frequently asked questions

Is retatrutide approved?

No. Retatrutide is an investigational medicine still in clinical trials. It is not approved by the US FDA or other major regulators. Your GP will explain what this means before any prescribing decision.

How is it different from semaglutide or tirzepatide?

Semaglutide acts on the GLP-1 receptor. Tirzepatide acts on GIP and GLP-1 receptors. Retatrutide is designed to act on GIP, GLP-1 and glucagon receptors, which is why it is called a triple agonist.

What formats are available and how should I store them?

GLP-3 RX is available as a pen or a premixed liquid vial. Both should be kept in the fridge at 2-8C, away from light and never frozen. Follow the storage instructions on your pharmacy label.

Are there side effects?

Side effects can occur. Your GP will review your health, medication and, where relevant, blood results before prescribing, and you should report anything unexpected.

How much should I take?

Your dose is set by our licensed GP following a consultation, and printed on your pharmacy label.
